The M16A1 is a modification of the XM16E1 (a modified variant of the original M16) to address problems found in the testing cycle of the XM16E1. Changes included a closed "birdcage" flash hider to replace the XM16E1's three-prong flash hider, which caught on twigs and leaves, a full fence around the magazine release to prevent accidental ejection, and a chrome plated chamber - later fully lined bore - to reduce fouling. The bolt was also changed so the cam pin could not be inserted with the bolt installed backwards, which would cause failures to eject until corrected. The front pivot point is also strengthened, giving the area in front of the magazine well a slightly different shape.

In the movie, Tony Montana (Al Pacino) uses a full auto-converted Colt AR-15 with an M203 grenade launcher attached during the final battle. In one of the film's more memorable moments, Tony yells out "Say hello to my little friend!" and blows a door down with a 40mm grenade, killing several of Alejandro Sosa's men. Also of note is that he has two magazines taped together 'jungle style'. After expending his first two magazines, he inserts a single 30-round magazine before the end. Originally an M16A1 was used with the grenade launcher, and the full auto-converted Colt AR-15 Sporter-1 was substituted after cinematographer John Alonzo determined that the AR-15’s firing sequence could be timed to synchronize with the Arriflex cameras, which would result in elongating the duration of muzzle flashes seen on film.

In the game, Scarface: The World Is Yours, an M16A1 with a strange vented handguard appears in this game as the "Carbine Assault Rifle". Like the AK-47, it can be fitted with "jungle style" dual magazines, and can also be fitted with an M203 grenade launcher. Tony uses one at the beginning of the game during the mansion shootout. The under-barrel grenade launcher appears to have been modelled after an actual M203 and not the mocked-up 39mm launcher prop used in the movie.

The gun is based on the real-life Colt M16A1 assault rifle which was used by the US Military starting in 1967 in Vietnam War, by 1969 it was the standard issue service rifle of the US Army. It is the second-best assault rifle in the game (with and without mods). Tony can purchase this gun for $2400 and carry 450 rounds or 900 rounds if Montana Defense is purchased; each magazine has 50 rounds, however, the dual magazine attachment mod can double the amount to 100 rounds. Tony can also buy an M203 and use it with M79 grenades (Tony can already use the jungle-style magazines in the mansion shootout, but he will lose them for the rest of the game, and the player must buy them if they intend to use them).
